EFFECT OF MMS AND CAPTAN ON DNA SYNTHESIS AND REPAIR SYNTHESIS IN DEVELOPING MOUSE EMBRYO CELLS

Abstract

|
|
Effects of MMS and captan on DNA synthesis and repair synthesis in early mouse
embryo cells were studied and the results obtained were as follows: 1. DNA synthesis:
One-tenth mM MMS severely inhibited the rates of DNA synthesis in both 2-cell stage
and morula stage cells to 83-86% of control at zero hour. This inhibition was further
enhanced with time, representing 41-48% at 2 hours, and 44-56% at 4 hours after
treatment, although this cencentration did not show any disturbance in DNA synthesis
in HeLa S©ýand CHO cells. These results indicated that early embryo cells are quite
sensitive to environmental mutagens. synthesis in both morula and blastocyst cells. Its
rate was dose-dependent and the maximum rates were 23-25% of total cells analyzed.
Captan (0.03-0.25 mM also induced unscheduled DNA synthesis and the maximum rates
were 11-13%. These results indicate that both repair damage induced in DNA by MMS
and captan, and that MMS may cause more damage than captan does.
